Sākotnēji šīs katoļu baznīcas vietā 1685. gadā par Krakovas karavadoņa Beļinska līdzekļiem tika uzcelta koka baznīca. Tā nodega negaisa laikā pēc diviem gadsimtiem – 1887. gadā. Jau nākošajā gadā nodegušās baznīcas vietā sākās jaunās katedrāles (no akmens) celtniecība. Arī mūsdienās šo katedrāli mēdz dēvēt par vienu no lielākajām un skaistākajām būvēm Latvijā. Projekta autors – Rīgas inženieris Florians Viganovskis. Katedrāles celtniecības laikā nomainījās vairāki dekāni un bija izteikts finansiālu līdzekļu trūkums, tādēļ ēkas beigu variantā varēja novērot dažas atkāpes no projekta pamatieceres.
Katedrāles apdares darbi ilga līdz pat 1902. gadam. Savukārt 1904. gadā Rēzeknes Jēzus Sirds katedrāle tika iesvētīta. Baznīcas formas atspoguļo sava laika arhitektūras iezīmes: fasādes daļa veidota pseidoromānikas, bet altārdaļa – pseidogotikas stilā.
Apmeklējot katedrāli, ir vērts pievērst uzmanību grieztajiem koka altāriem, kas rotāti ar Jēzus Kristus, Jaunavas Marijas, Svētās Terēzes un citu svēto skulptūrām. Katedrāle ir slavena ar savām vitrāžām un pirmo Livonijas bīskapu – Svētā Meinarda un Svētā Alberta, attēlojumiem tajās.
Kopš 1995. gada te atrodas Rēzeknes-Aglonas diecēzes kūrija.
