Baznīcas dibināšanas akts apstiprināts 1567. gadā un tā ir vecākā līdz mūsdienām saglabājusies Jelgavas celtne, kā arī pirmā luterāņu baznīca Latvijā, kas atjaunota pēc II Pasaules kara.
Jelgavas Sv. Annas prokatedrāle ir viens no retajiem manierisma stila pieminekļiem Latvijā. Baznīcas gotiskie elementi ir tornis ar smaili un augstie logi. Prokatedrāle apskatāma 1910. gadā gleznotā Jāņa Rozentāla glezna “Kristus un samariete” 1994.g.gadā baznīcā tika uzstādītas Gēteborgas meistara Olafa Hammarberga 1938.gadā būvētās ērģeles. Tām ir 37 reģistri un 2072 stabules. Ar savu vareno skanējumu ērģeles kalpo ne tikai draudzes vajadzībām, bet arī pulcē daudzus garīgās mūzikas cienītājus. Dievnamā regulāri notiek garīgās un klasiskās mūzikas koncerti, kuros uzstājas augsta līmeņa Latvijas un ārvalstu izpildītāji.
